5172 CTSA Program Scholar Receives Prestigious NIH Grant-1 Clinical and Translational Science Awards (CTSA) Program scholar Ying Liang, M.D., Ph.D., an assistant professor of toxicology and cancer biology at the University of Kentucky (UK), recently received her first NIH Research Project Grant (R01). The R01 grant will fund her research on the role of a protein called latexin in preventing cancer therapy-induced stem cell damage. The 2015 applicant success rate for R01-equivalent NIH grants was just 18.9 percent, and Liang credits NCATS’ CTSA Program training support ― known as a KL2 award ― as being instrumental in overcoming the odds. UK’s Center for Clinical and Translational Science faculty members selected her as a KL2 scholar in 2012, and with those funds, she set out to identify how latexin makes blood and stem cells more susceptible to damage induced by cancer therapy. CTSA Program scholar support enabled Ying Liang, M.D., Ph.D., to obtain her first NIH Research Project Grant (R01) and continue her research on preventing chemotherapy-induced cell damage. (University of Kentucky photo) “In addition to the KL2 funding for my research, I received guidance from a multidisciplinary team of research and career development mentors and coursework on research methods, protocol development and grant writing,” Liang said. Chemotherapy and radiation target cancer by killing rapidly dividing tumor cells; however, they also damage rapidly dividing healthy cells and stem cells in the bone marrow. This damage can cause bleeding problems and increase the risks of infection and secondary cancers. Liang and her team found that mice that carried bone marrow stem cells genetically engineered to make less latexin had much lower rates of radiation-induced complications and death than control mice. The findings suggested that latexin could be a potential target for treatments that help prevent radiation-related stem cell damage. Providing the resources to train, cultivate and sustain future leaders of the translational research workforce is a key NCATS goal and a major emphasis for the CTSA Program. Most CTSA Program hubs have a Clinical Research Scholar (KL2) program for those who have a doctoral degree in medicine or the sciences. KL2 awards support mentored career development for investigators who have recently completed professional training and are just starting out in their research careers. With the R01 support, Liang and her team will continue to tease apart the mechanisms by which latexin makes stem cells vulnerable to cancer therapy. 5146 SMR Key Facts & Questions Definitions A Molecular Libraries Screening Center Network (MLSCN) probe, which was originally part of the first phase of the Molecular Libraries Program (MLP), is a compound that represents an improvement over the existing art. Supporting information is required, showing currently available probes, their properties and how the new probe is clearly an improvement (first or best in its class). Potency: Criteria for potency are context dependent, varying with the assay and target type and the current state of the art. In general, probes identified via biochemical assays are expected to exhibit potencies of less than 500 nanomolars (nM) and more ideally in the 100-nM range. Those identified in cell-based assays are expected to exhibit potencies at a level of less than 1 micromolar (μM). However, in certain instances, potencies in the 10-μM range may be acceptable. Criteria for whole-organism screening are less stringent. Solubility: Sufficient solubility in relevant solvents. Availability: The probe molecule should be accessible in amounts to allow advanced studies (15 milligrams [mg] to 20 mg), and protocols for its preparation or isolation should be made available. Structure-activity relationship, mode of action (e.g., evidence of binding to target, characterization of mechanism of action) and awareness of selectivity against relevant and/or related targets are expected. 5145 SMR Compound Requests Requests for compound sets from the NCATS Small Molecule Resource (SMR) should be done through the Evotec-NCATS SMR Compound Management website. When the compound order is registered at the website, the requester will receive a standard Material Transfer Agreement (MTA) form that defines an understanding of the investigators responsibilities in receiving the ordered compounds. The requester will need to sign the MTA and send it to Evotec before the compound set(s) will be sent.
5144 SMR Data Sharing Structure information for all compounds in the NCATS Small Molecule Resource (SMR) and all data generated through testing of the SMR compounds—which will include but not be limited to assay descriptions, assay protocols and/or links to published assays; data from measurements of compound chemical and physical properties; performance data for assays and compounds; and primary data from high-throughput screening and data generated in the secondary screen (e.g., EC50s, IC50s, AC50s, counter screens)—will be deposited into PubChem, a public database, where data will be available to all researchers, in both the public and private sectors, for further use in studying biology and disease. Consistent with the goals of the SMR, data generated through screening of the SMR will be made available to the public within 2 years of receipt of SMR compounds, either through publications or through deposit in PubChem or other NIH-approved databases.
5143 SMR in Action News A probe project executed by The Scripps Research Institute (TSRI), using compounds from the NCATS Small Molecule Resource (SMR), has become the program’s first to yield a drug candidate tested in humans. The initial probe compound resulting from screening an early version of the NCATS SMR collection was found to be an agonist of the sphingosine-1-phosphate receptor 1 (S1P1). The active compounds and related molecules were further developed by Receptos, Inc., eventually resulting in administration to the first human subject in a Food and Drug Administration-approved clinical trial for multiple sclerosis (MS) in 2011. Now named RPC1063, the drug candidate successfully completed Phase IIB clinical trials in November 2014 and recently began Phase III.  MS is a neurodegenerative disease that affects the central nervous system (brain and spinal cord). Patients with MS suffer damages to myelin, the material that surrounds and insulates nerve cells. There are approximately 250,000 to 350,000 people in the United States with MS diagnosed by a physician. MS is currently believed to be an immune-mediated disorder. S1P1 plays a key role in the immune system, regulating lymphocyte egress from lymphoid tissues into the circulation. It is believed that S1P1 agonists induce T-cell sequestration in lymph nodes, diminishing their ability to reach distant sites and contribute to MS pathology, including inflammation and demyelination. SAN DIEGO, Dec. 22, 2014 (GLOBE NEWSWIRE) — Receptos, Inc. (NASDAQ: RCPT) announced today that the company has enrolled the first patient in the SUNBEAM Phase III trial of RPC1063 in patients with relapsing multiple sclerosis (RMS). The company had previously initiated the Phase III portion of the RADIANCE trial in patients with RMS. With both trials now actively enrolling patients, Receptos is on track to complete this Phase III clinical development program in 2017. “Our initiation of the SUNBEAM trial represents another major corporate milestone for Receptos and positions RPC1063 as the potential next-to-market oral agent for the treatment of relapsing multiple sclerosis,” said Faheem Hasnain, president and chief executive officer of Receptos. “Following our recent positive Phase II results in ulcerative colitis, we also plan to initiate a Phase III program for RPC1063 in inflammatory bowel disease in 2015. We believe that this drug candidate represents a potential franchise in immunology, and we continue to explore opportunities in other therapeutic indications where there is strong scientific rationale.” BURLINGAME, Calif., Jan. 6, 2015 — Cleave Biosciences today announced that it has begun a Phase I clinical trial to evaluate its lead drug candidate, CB-5083, in patients with advanced solid tumors. CB-5083 is a first-in-class, oral inhibitor of p97, a critical enzyme that controls various aspects of protein homeostasis. P97 inhibition is a novel approach that has the potential to treat a wide range of cancers, including both solid tumors and hematologic malignancies. CB-5083 is also being studied in a Phase I trial in patients with relapsed and refractory multiple myeloma. CB-5083 is derived from a compound initially synthesized by the Specialized Chemistry Center at the University of Kansas, which is part of the National Institutes of Health’s Molecular Libraries Program (which found an active compound while screening the NCATS SMR collection). Cleave’s discovery partners include Tsui-Fen Chou, Ph.D., from the laboratory of Raymond J. Deshaies, Ph.D., who is a professor of biology at the California Institute of Technology, a Howard Hughes Medical Institute investigator and a scientific founder of Cleave Biosciences; and TSRI, also a member of the Molecular Libraries Program.
5142 SMR Operations The major operations of the NCATS Small Molecule Resource (SMR) include: Identifying and acquiring small molecule compounds; Applying a standard quality control (QC) regimen; Handling, storing and maintaining the compound collection within established guidelines; Distributing compound plates to the research community and Depositing compound structures into PubChem.   Identification and Acquisition of Compounds An NCATS medicinal chemist works to optimize a small molecule probe. (NCATS) The SMR compound collection began as part of the Molecular Libraries Probe (MLP) Production Centers Network (MLPCN). The collection was initially developed to supply the Network with low-molecular-weight compounds for high-throughput screening (HTS). In coordination with the External Scientific Advisory Committee of the MLPCN, the SMR developed compound selection criteria (e.g., stability, solubility, availability of sufficient quantities, ease of re-supply or re-synthesis, diversity). Compounds were obtained in several different ways, including purchase from commercial vendors, grants to medicinal and organic chemists and donations from academic investigators. Compound Availability The SMR is currently offering compound sets selected for known biological activity against specific protein targets or signaling pathways. The NIH Clinical Collection set is a plated array of approximately 700 small molecules that have a history of use in human clinical trials. Similar collections of Food and Drug Administration-approved drugs have proven to be rich sources of undiscovered bioactivity and therapeutic potential. Soon to be released, the Probe Plus set is a collection of 1,400 compounds containing probes discovered through the screening effort of the MLP. For each probe, the set contains five structural analogs that will provide important structure activity information from each screen. QC The SMR processes all samples that it has received in a standard, high-throughput QC regimen that checks for weight, solubility, identity and purity. Sample weight is verified to ensure that the minimum amount required was received. Sample solubility in dimethyl sulfoxide (DMSO) is checked, and only completely soluble samples are accepted into the SMR. Liquid chromatography-mass spectrometry is used to determine sample identity and purity. The SMR accepts samples where the molecular ion is identified and the area under the curve is at least 90 percent by evaporative light scattering or ultraviolet detection. Compound Handling, Storage and Distribution The SMR immediately stores samples that pass QC tests in automated storage equipment for fast retrieval. SMR uses a three-tier storage system: long-term store, working store, and short-term store. The SMR stores the bulk sample dry in glass vials as the long-term store. Working-store samples of each compound are created in a 10-micromolar DMSO solution to be stored and used for approximately one year. The working-store samples are in turn used to create microtiter plates to be shipped to investigators, which are then held in the short-term store until shipment (typically less than one week). Compounds are stored at -20°C in all three tiers of the storage system, and all DMSO solutions are stored and handled under nitrogen atmosphere to prevent water absorption and ensure optimal stability and purity.
